In a significant leap for sustainable food production, Israeli startup DairyX Foods has unveiled a groundbreaking method to produce cow-free casein proteins using precision fermentation. This innovation, which enables the creation of self-assembling casein micelles, promises to deliver authentic dairy products like cheese and yogurt without relying on traditional animal-based processes, marking a pivotal shift in the alternative dairy landscape.

“We’ve designed our strain to produce casein that not only matches the amino acid sequence of milk casein, but also carries the same PTMs, which are crucial for its self-assembly and performance, just as it does in milk,” founder and CEO, Arik Ryvkin told.

Revolutionizing Dairy with Animal-Free Casein

For decades, the dairy industry has depended on bovine-derived casein, the protein responsible for 80% of milk’s protein content and critical for properties like emulsification, thickening, and foaming. There are four kinds of casein proteins in cow’s milk, which self-organise into micelles. These are highly hydrated spherical structures that serve as a building block for the functional and nutritional attributes of dairy products

Casein proteins usually undergo post-translational modification, particularly phosphorylation, or the addition of phosphate groups, which directly interact with calcium to help form the micelles. When produced with microbial systems, casein proteins require precise phosphorylation to retain their functional properties.

“We have achieved several key objectives with our solutions. Our biological design genetically manipulated yeast to produce functional caseins that we organised into micelles. We developed a fast-tracked screening process that simulated evolution to locate super-producers of protein from among millions of yeast strains,” outlines Kuznets.

“Our machine-learning models simulated fermentation to determine optimal fermentation conditions. We have proven our ability to create a gel from reconstituted casein micelles. All these ingenuities have helped us work smarter and faster to create highly functional micelles.”

The startup’s commitment to replicating bovine casein's post-translational modifications ensures that its “smart casein” delivers the same gelation and emulsification properties as traditional dairy, making it compatible with existing manufacturing processes. This functionality is critical for producing firm, stretchy, and creamy dairy products that meet consumer expectations.

DairyX is already engaging with potential partners to co-develop technology and products, with a commercial launch targeted for 2027. “We expect our first product using our casein to hit the market in 2027,” suggests Ryvkin.
