KAIST Team Unveils Pioneering AI Design Technology

Researchers from the Korea Advanced Institute of Science and Technology (KAIST) have announced a significant leap in personalized cancer therapy with the development of a novel AI model designed to create more effective B cell-based cancer vaccines. This technology directly targets a major limitation in current immunotherapy approaches.

Led by Professor Jung Kyoon Choi from the Department of Bio and Brain Engineering, in collaboration with Neogen Logic Co., Ltd., the team has created the first AI technology capable of quantitatively predicting B cell reactivity to tumor-specific markers called neoantigens. This advancement moves the field beyond a sole focus on T-cell responses, aiming to generate longer-lasting immune memory against cancer.

Overcoming a Critical Immunotherapy Gap

Current next-generation cancer vaccine development, pursued by companies like Moderna and BioNTech, has primarily concentrated on stimulating T cells. This approach, while promising, often overlooks the critical role of B cells, which are responsible for producing targeted antibodies and establishing the immune system's long-term memory.

The KAIST team's work addresses a concern recently highlighted by oncologists at Johns Hopkins University. In a May 2025 commentary in Nature Reviews Cancer, Professors Mark Yarchoan and Elizabeth Jaffee noted, "Despite accumulating evidence regarding the role of B cells in tumor immunity, most cancer vaccine clinical trials still focus only on T cell responses."

The Core of the Innovation: An Integrative AI Model

The newly developed AI model overcomes this gap by learning the structural binding patterns between mutated cancer proteins and B cell receptors (BCR). It integrally analyzes both T cell and B cell reactivity to neoantigens, unique protein fragments derived from a patient's specific cancer mutations.

"By adding B cell reactivity, cancer vaccines can move beyond one-time attacks and short-term memory to become a long-term immunity that 'remembers' cancer, effectively preventing recurrence," explained the KAIST announcement.

"The KAIST team's work addresses a concern recently highlighted by oncologists at Johns Hopkins University. This technology has been validated through extensive analysis of large-scale cancer genome data, animal experiments, and existing clinical trial data, confirming that integrated B cell responses significantly enhance anti-tumor effects.

Pathway to Clinical Application

Professor Choi confirmed the translational path for this discovery, stating, "Together with Neogen Logic Co., Ltd., which is currently commercializing neoantigen AI technology, we are conducting pre-clinical development of a personalized cancer vaccine platform and are preparing to submit an FDA IND with the goal of entering clinical trials in 2027."

An FDA IND (Investigational New Drug) application is the necessary regulatory step to begin human trials.

The research, with Dr. Jeong Yeon Kim and Dr. Jin Hyeon An as co-first authors, was published in the peer-reviewed journal Science Advances on December 3. The study, titled "B cell–reactive neoantigens boost antitumor immunity," represents a foundational step toward more comprehensive and durable personalized cancer vaccines.
